The effect of mode of incorporation of superdisintegrants like croscarmellose sodium, sodium starch glycolate and crospovidone (polyplasdone XL and XL-10) on dissolution of three model drugs with varying aqueous solubility, like carbamazepine (poorly soluble), acetaminophen (sparingly soluble) and cetrizine HCl (soluble) from their respective tablet formulations prepared by wet granulation was studied. The disintegrants were incorporated extragranularly or intragranularly or distributed equally between the two phases. The results indicated that Crospovidone in general was effective in improving the dissolution of the drugs used in the study and generally extragranular mode of addition seemed to be the best mode of incorporation, irrespective of the solubility of the main tablet component.
